Las c\u00e9lulas que expresan un GPCR de inter\u00e9s que emite se\u00f1ales a trav\u00e9s del calcio est\u00e1n precargadas con Fura-2 AM que puede atravesar la membrana celular. Una vez dentro de la c\u00e9lula, los grupos bloqueadores lipof\u00edlicos de Fura-2 AM son escindidos por las esterasas, lo que da como resultado un tinte fluorescente cargado negativamente que permanece dentro de las c\u00e9lulas y su longitud de onda de fluorescencia cambia al azul al unirse al calcio. Cuando las c\u00e9lulas se estimulan con agonistas, el receptor se\u00f1ala la liberaci\u00f3n de calcio intracelular, lo que aumenta en gran medida la intensidad de fluorescencia de Fura-2 en la longitud de onda corta. <\/p>\n\n\n\n<p>Las caracter\u00edsticas radiom\u00e9tricas de Fura-2 hacen de este kit una herramienta ideal para una medici\u00f3n m\u00e1s precisa de la concentraci\u00f3n de calcio celular en comparaci\u00f3n con Fluo-4 de longitud de onda \u00fanica. Soluci\u00f3n madre Fura-2 AM <\/em><\/p>\n\n\n\n<p style=\"font-size:16px\">Agregue 200 \u00b5L de DMSO en el vial de Fura-2 AM (Componente A) y m\u00e9zclelos bien. Tenga en cuenta que 20 \u00b5L de soluci\u00f3n madre de Fura-2 AM es suficiente para una placa. La soluci\u00f3n madre de Fura-2 AM no utilizada puede dividirse en al\u00edcuotas y almacenarse a &lt; -20 oC durante m\u00e1s de un mes si los tubos est\u00e1n sellados herm\u00e9ticamente. Proteger de la luz y evitar ciclos repetidos de congelaci\u00f3n-descongelaci\u00f3n.  <\/p>\n\n\n\n<p style=\"font-size:16px\"><em>2. Buffer de ensayo (1X) <\/em><\/p>\n\n\n\n<p style=\"font-size:16px\">Para el Cat. 36320 (kit de 10 placas), prepare buffer de ensayo 1X agregando 9 ml de HHBS (componente C) en Pluronic\u00ae F127 Plus 10X (1 ml, componente B) y m\u00e9zclelos bien). Para el Cat. 36321 (kit de 100 placas), prepare buffer de ensayo 1X agregando 90 ml de HHBS (no incluido) en Pluronic\u00ae F127 Plus 10X (10 ml, componente B) y m\u00e9zclelos bien. Tenga en cuenta que 10 ml de buffer de ensayo 1X es suficiente para una placa. Haga al\u00edcuotas y almacene el buffer de ensayo 1X no utilizado a &lt;\u00a0-20\u00a0<sup>o<\/sup>C.
