Cultive las c\u00e9lulas a una densidad \u00f3ptima para obtener im\u00e1genes de acuerdo con su protocolo de inducci\u00f3n espec\u00edfico (alrededor de 1 &#8211; 2 \u00d7 104 c\u00e9lulas\/pocillo\/placa de 96 pocillos).<\/p>\n\n\n\n<p style=\"font-size:16px\">2. Para c\u00e9lulas vivas: Incube las c\u00e9lulas con StrandBrite\u2122 RNA Green (Componente A) diluido 400X en medio o buffer de tinci\u00f3n de c\u00e9lulas vivas (Componente B) a temperatura ambiente durante 30 a 60 minutos (100 \u00b5L\/pocillo). <strong>Nota:<\/strong> 25 \u00b5L de StrandBrite\u2122 RNA Green (Componente A) son suficientes para una placa de 96 pocillos. Proteger de la luz y evitar ciclos repetidos de congelaci\u00f3n-descongelaci\u00f3n. El tiempo de incubaci\u00f3n adecuado depende del tipo de c\u00e9lula individual y de la concentraci\u00f3n celular utilizada. Optimice el tiempo de incubaci\u00f3n para cada experimento. Para c\u00e9lulas fijadas: fije las c\u00e9lulas con metanol puro durante 1 minuto a temperatura ambiente y luego lave con PBS. Sumerja las c\u00e9lulas en Triton-100 al 1% durante 2 minutos, luego lave dos veces con PBS. Incube las c\u00e9lulas con StrandBrite\u2122 RNA Green (Componente A) a la concentraci\u00f3n de 1X en PBS a temperatura ambiente durante 15 a 30 minutos. <strong>Nota: <\/strong>Se recomienda aumentar la concentraci\u00f3n de marcaje o el tiempo de incubaci\u00f3n para permitir que se acumule el tinte si las c\u00e9lulas no parecen estar lo suficientemente te\u00f1idas.
